Highly sensitive bacterial susceptibility test against penicillin using
parylene-matrix chip

This workpresentedahighlysensitivebacterialantibioticsusceptibilitytestthrough β-lactamase assay using Parylene-matrixchip. β-lactamases (EC3.5.2.6)areanimportantfamilyofenzymesthatconfer resistance to β-lactam antibioticsbycatalyzingthehydrolysisoftheseantibiotics.Herewepresenta highly sensitiveassaytoquantitate β-lactamase-mediated hydrolysisofpenicillinintopenicilloicacid. Typically,MALDI-TOFmassspectrometryhasbeenusedtoquantitatelowmolecularweightanalytesand to discriminatethemfromnoisepeaksofmatrixfragmentsthatoccuratlow m/z ratios (m/zo500).The β-lactamase assayforthe Escherichiacoli antibioticsusceptibilitytestwascarriedoutusingParylene- matrix chipandMALDI-TOFmassspectrometry.TheParylene-matrixchipwassuccessfullyusedto quantitatepenicillin(m/z: [PENþH]þ¼335.1and[PENþNa]þ ¼357.8)andpenicilloicacid(m/z: [PAþH]þ¼353.1)ina β-lactamase assaywithminimalinterferenceoflowmolecularweightnoise peaks.The β-lactamase assaywascarriedoutwithanantibiotic-resistant E. coli strain andanantibiotic- susceptible E. coli strain, revealingthattheminimumnumberof E. coli cells requiredtoscreenforan- tibiotic resistancewas1000cellsfortheMALDI-TOFmassspectrometry/Parylene-matrixchipassay.
